HEMATOPOIETIC II LABORATORY


Carefully examine the slides listed below. For this lab, histologic sections of a human embryo and bone marrow will be examined. In addition, you must examine the bone marrow aspirate carefully to find members of the granulocytic and erythroid cell lines.

Slide #91, bone marrow aspirate smears:

  1. The bone marrow aspirate is obtained by inserting a needle through the skin subcutaneous tissues and cortical bone and withdrawing by aspiration of cylindrical mass of red marrow. (Yes, Virginia, this process is very painfull) The aspirate is then smeared on a slide and prepared for microscopic viewing.
  2. Examine the smear under low power (10X). Determine the distribution of component cells and nests of hematopoiesis. Using oil immersion (use the lens with the black ring!), identify members of theerythropoietic series from basophilic normoblast through erythrocyte. Identify members of the granulocyticseries from promyelocyte through neutrophil, basophil and eosinophil. Identify a blast cell and a memberof the megakaryocyte series. How can you distinguish a megakaryocyte fro an osteoclast?
